Thermocycler Calibration and Validation

Thermocyclers are instruments used to amplify DNA and RNA samples by the polymerase chain reaction. The thermocycler raises and lowers the temperature of the samples in a holding block in discrete, pre-programmed steps, allowing for denaturation and reannealing of samples with various reagents. Amplified genetic material can be used in many downstream applications such as cloning, sequencing, expression analysis, and genotyping.

To ensure your thermocycler raises and lowering temperature accurately, we perform the following calibrations:

We check the temperature at 24 wells in 96-wells at the denaturation temperature of 92-95 Celsius.

We check the temperature of 24 wells in 96-wells block at the annealing stage of 50-55 Celsius.

We check the temperature of 24 wells in 96-wells block at the elongation stage of 65-70 Celsius.
